That's odd...
I just experimented a bit with my own, and the change took effect immediately. I am running on-premises virtual edges, though...haven't tried with the AWS/BYOC-Cloud edges.
What is your configuration?

I am pretty sure if you add it to the queue settings it takes a while to refresh..not hours, but certainly not instant

I've been having some problems getting queue whisper prompts to refresh. Are these cached? I've tried re-uploading audio, removing the prompt from the queue config and adding it back in, but it is still playing an "old" version of the prompt even an hour later.
